Abstract: PURPOSE: To analyze the literature on epigenetic regulatory mechanisms of DNA methyltransferase 3B (Dnmt3B) in osteoarthritis (OA), delineate the research landscape, key trends, and limitations in this field, and provide insights for future studies. METHODS: Original publications published between 2010 and 2025 were retrieved from PubMed, Web of Science, Scopus, and CNKI databases. CiteSpace 6.3.R1 was employed for keyword co-occurrence analysis, clustering, prominence analysis, and bibliometric visualization. RESULTS: A total of 21 valid articles were identified. Research focuses primarily revolve around DNA methylation, chondrocytes, and inflammation, with an increasing emphasis shifting from fundamental mechanism elucidation to the analysis of Dnmt3B-specific signaling pathways. Dnmt3B is involved in maintaining cartilage homeostasis, inflammatory responses, and metabolic reprogramming through regulation of pathways such as miR-34a, ABAT, and Wnt/β-catenin. Interdisciplinary approaches incorporating natural compounds, animal models, and metabolic epigenetics represent emerging research directions; however, existing studies predominantly concentrate on single pathways and basic experiments, lacking sufficient clinical translational evidence. CONCLUSIONS: Dnmt3B serves as a pivotal molecular player in OA epigenetic regulation, with its aberrant expression driving cartilage degeneration, bone remodeling, and synovial inflammation. Future investigations should integrate multi-omics data, conduct large-scale clinical cohort studies, and develop precision intervention strategies, ultimately establishing diagnostic biomarker systems based on Dnmt3B genetic polymorphisms and methylation profiles.
