Relationship between expression of hypoxia-inducible factor 1α and clinical characteristic of recurrent ameloblastoma

摘要: 目的:检测缺氧诱导因子1α(hypoxia-inducible factor 1α,HIF-1α)在成釉细胞瘤中的表达,并探讨其与成釉细胞瘤侵袭和复发的相关性。方法:对5对成釉细胞瘤及对应正常瘤旁组织进行转录组测序,并对60例原发及60例复发患者第1次手术标本行免疫组织化学染色,检测2组患者中HIF-1α蛋白的表达。采用SAS9.3软件包对数据进行统计学分析。结果:5对新鲜标本转录组测序发现,3对标本中HIF-1α及其相关分子转录水平呈明显一致性改变。免疫组织化学染色发现,复发组成釉细胞瘤中,40%的肿瘤HIF-1α染色高于未复发组;复发组中,48.33%的肿瘤slug染色高于未复发组,差异具有统计学意义(P<0.05)。结论:成釉细胞瘤复发与HIF-1α相关,HIF-1α表达高的患者较表达低的患者更容易复发。HIF-1α可能通过调控slug的表达,影响成釉细胞瘤的局部复发。

Abstract: PURPOSE:To detect the expression of hypoxia-inducible factor 1α in ameloblastoma and to determine if it was involved in the recurrence of ameloblastoma. METHODS: Five pairs of ameloblastoma and normal para-tissue were used for transcriptional group sequence; 120 formalin-fixed, paraffin-embeded tissue blocks of ameloblastoma specimens were compared at protein level using immunohistochemistry for HIF-1α staining. The data were analyzed with SAS 9.3 software package. RESULTS: The transcriptional level of HIF-1α and its related molecules in 3/5 specimens were changing consistently and obviously after sequencing; the HIF-1α and slug expression levels were significantly higher in recurrent ameloblastoma. About 40% of recurrent ameloblastoma had higher HIF-1α staining than primary ameloblastoma and 48.33% of recurrent ameloblastoma had higher slug staining than primary ameloblastoma (P<0.05). CONCLUSIONS: Higher expression of HIF-1α plays an important role in the recurrence of ameloblastoma.
